Application of nucleic acid based techniques for detection of Classical swine fever virus: A comparative study
نویسندگان
چکیده
Classical swine fever (CSF) is a contagious and devastating viral disease, causing serious losses in the pig industry worldwide. In the control programmes of CSF, rapid detection and identification of the causative agent is a crucial step. Various PCR based techniques like nested RT-PCR, SYBRGreen based RealTime PCR and TaqMan based Real-time PCR were used for detection of CSFV nucleic acid in clinical as well as tissue samples. In our study three detection systems were tested for classical swine fever virus (CSFV) detection and for its discrimination from other pestiviruses; Nested PCR, non-specific dsDNA-binding SYBR Green dye based and specific fluorogenic TaqMan MGB probe based Real-Time PCR. However, one-step TaqMan Real-Time PCR assay was shown to be the most appropriate for pestivirus discrimination in comparison to the other two assays, moreover, it reduces the risk of contamination and is less time consuming.

Rope-based oral fluid sampling for early detection of classical swine fever in domestic pigs at group level
BACKGROUND Non-invasive sampling techniques based on the analysis of oral fluid specimen have gained substantial importance in the field of swine herd management. Methodological advances have a focus on endemic viral diseases in commercial pig production. More recently, these approaches have been adapted to non-invasive sampling of wild boar for transboundary animal disease detection for which ...
متن کاملEvaluation of Nucleic Acid Sequence Based Amplification (NASBA) and Reverse Transcription Polymerase Chain Reaction for Detection of Coxsackievirus B3 in Cell Culture and Animal Tissue Samples
Enteroviruses are the causative agents of a number of diseases in humans. Group B coxsackieviruses are believed to be the most common viral agents responsible for human heart disease. Genomic data of enteroviruses has allowed developing new molecular approaches such as Nucleic Acid Sequence Based Amplification (NASBA) for detection of such viruses. In this study, coxsackievirus B3 (CVB3) was de...
متن کاملMolecular Detection of Torque Teno Sus Virus and Coinfection with African Swine Fever Virus in Blood Samples of Pigs from Some Slaughterhouses in Nigeria
Torque teno sus virus 1 (TTSuV1a/TTSuV1b) infection is present in pig herds worldwide. This study investigated the prevalence of TTSuV1a/TTSuV1b infections in domestic pigs from some slaughterhouses in Nigeria as well as coinfection with African swine fever virus (ASFV) and described the phylogeny in relation to global strains. One hundred and eighty-one (181) blood samples from four slaughterh...
متن کاملPreclinical Detection of Porcine Circovirus Type 2 Infection Using an Ultrasensitive Nanoparticle DNA Probe-Based PCR Assay
Porcine circovirus type 2 (PCV2) has emerged as one of the most important pathogens affecting swine production globally. Preclinical identification of PCV2 is very important for effective prophylaxis of PCV2-associated diseases. In this study, we developed an ultrasensitive nanoparticle DNA probe-based PCR assay (UNDP-PCR) for PCV2 detection. Magnetic microparticles coated with PCV2 specific DN...
متن کاملVisual detection and differentiation of Classic Swine Fever Virus strains using nucleic acid sequence-based amplification (NASBA) and G-quadruplex DNAzyme assay
The split G-quadruplex DNAzyme has emerged as a valuable tool for visual DNA detection. Here, we successfully integrated colorimetric split G-quadruplex DNAzyme assay with nucleic acid sequence-based amplification to generate a novel detection approach, allowing visual and rapid detection for the RNA of Shimen and HCLV strains of Classic Swine Fever Virus (CSFV). CSFV is a RNA virus that causes...
